New Study on Psilocybin Reveals Disruptions in Brain Networks

August 2, 2024 — Psilocybin the active ingredient in magic mushrooms, has shown promise in treating various mental health conditions, but the specifics of its impact on brain communication networks remained unclear until now.

Researchers at the Washington University School of Medicine in St. Louis conducted an in-depth study, recently published in Nature, to explore the effects of psilocybin on the brain’s functional networks. The study involved scanning the brains of seven healthy participants multiple times before, during, and after ingesting psilocybin.

Groundbreaking Brain Scans

The participants underwent an average of 18 brain scans each using functional magnetic resonance imaging (fMRI), which measures blood flow changes in the brain. The results revealed that psilocybin disrupted connections within established brain networks while increasing communication between different networks. This indicates that psilocybin makes the brain’s information processing more unpredictable and disordered.

Notably, the researchers observed lasting brain changes in the days and weeks following the psychedelic experience. These changes were particularly evident in the connections between the hippocampus, associated with short-term memory, and the default mode network, active when a person is at rest. These findings suggest that psilocybin may enhance the brain’s neuroplasticity and could explain its therapeutic effects in treating anxiety, depression, and addiction.

Limitations and Concerns

However, the study’s findings are primarily based on a small sample of six participants, as one participant dropped out. This small sample size and the lack of information about the participants’ prior experiences with psychedelics raise concerns about selection bias and the generalizability of the results.

Additionally, while the study used an active control (a stimulant drug) to mitigate placebo effects, it did not address whether participants and researchers could distinguish between psilocybin and the placebo during the experiment. This is a common challenge in psychedelic studies, as the psychoactive effects of psilocybin make it difficult to maintain a double-blind procedure. Consequently, it remains unclear whether the observed brain activity changes are solely due to psilocybin or also influenced by participants’ expectations.

Conflicts of Interest and Protocol Deviations

Many authors of the study report conflicts of interest related to the commercialization of neurotechnologies used in the research. The paper does not clearly outline how potential biases were mitigated. Furthermore, there appear to be deviations from the study protocol, such as the lack of reported data on long-lasting changes in participants’ well-being, measured by the persisting effects questionnaire. This data could have provided insights into the clinical relevance of psilocybin.

While the changes in brain patterns observed in the study are striking, it is unclear what these changes mean for participants’ subjective experiences. To fully understand the impact of psilocybin, researchers need to integrate objective brain data with subjective self-reported data.

Crucial questions remain: What does it mean for a person to have a more disordered brain under psilocybin? How do these changes in brain activity relate to how individuals feel and function in their daily lives?

This study represents a significant step in understanding psilocybin’s impact on the brain, but more research is needed. By combining neuroscience with methods that bridge the gap between objective brain data and subjective human experience, researchers hope to clarify whether the observed brain changes translate into meaningful therapeutic benefits. Until then, it is important to temper expectations for psilocybin’s potential based on these initial findings.
